Fastest to rev up from idle to redline? Probably Lotus.
Fastest revving in normal driving? Probably some Honda 4 cylinder.
Engine in legal road car with 'fastest' top speed? Most likely Bugatti's W 16.
Fastest 0-60mph? Check out C & D or R & T.
Fastest 1/4 mile drag motor? Some huge custom-made (not Dodge) blown hemi-V8 fueler.
Fastest still running at end of 24 Hours of LeMans? Audi's killer diesels!
Fastest in my front driveway? Azera 3.8l V6! (Back driveway has several faster projects!)
'Fastest' is not a really precise term, by itself.

Less than a two months ago a small British car company represented its version of ultimate race car for the road drive based on Chevrolet V8 engine which provides 730 hp and entered the Guinness Book of World Records beating the Veyron’s time by five hundredths of a second. 720 GTR scorches from 0 to 60 mph in 2.6 seconds, from 0 to 100 mph in 5.3 seconds, and brakes to 0 in 3.6 seconds (not including drivers reaction time). No traction control devices or ABS braking aids were used.
I have a 2007 Azera with a 3.8 engine.
